不同浓度IL-17A中和性抗体对CIA小鼠血清中炎症细胞因子表达的影响*

      [摘要]目的 通过建立胶原诱导关节炎(CIA)小鼠模型,探讨不同浓度的IL-17A中和性抗体对CIA小鼠血清中炎症细胞因子表达。方法 将48只SPF级C57BL/6雄性小鼠随机分为4组:A(空白对照)组、B(CIA+生理盐水)组、C(CIA+高浓度)组、D(CIA+低浓度)组;采用鸡II型胶原加氟氏完全佐剂乳化制备抗原尾部多点皮下注射;初次免疫2周后以同样方法加强免疫,诱导CIA小鼠模型出现后在小鼠腹腔注射细胞因子中和性抗体或生理盐水,观察2周并记录小鼠关节及关节外表现,检测血清TNF-a、IL-17、IL-6表达水平。结果C、D与B比较TNF-a、IL-17、IL-6表达水平显著降低,并且IL-17A中和性抗体高浓度组较低浓度组血清中炎症细胞因子表达水平降低,差异有统计学意义(P<0.05)。结论IL-17A中和性抗体可显著降低CIA小鼠模型血清中炎症细胞因子TNF-a、IL-17、IL-6表达水平,且以高浓度组较低浓度组改善骨关节功能更显著,其作用机制可能是通过调节Th17细胞参与介导的细胞因子-破骨细胞损伤免疫机制,抑制炎症细胞因子表达,进一步影响破骨细胞因子来实现。

      [Abstract] Objective The establishment of collagen induced arthritis model, Discussion different concentrations of IL-17A neutralizing antibody to CIA serum inflammatory cytokines in mice. Methods 48 male SPF C57BL/6 mice were randomly divided into 4 groups: A(Normal group)group; B(The CIA and Saline Group)group; C(The CIA and high concentration group)group; D(The CIA and low concentration group)group. Use of chicken type II collagen fluorine's complete adjuvant emulsion preparation of antigen tail more subcutaneous injection; Two weeks later, Primary immune with the same method for the second immunization, intraperitoneal inject neutralizing anti -murine interleukin-17 antibody or physiological saline after CIA mice models were induced; persistent observation and data record of the change of the general situation, mice weight,arthritis index and thickness of fat in two weeks, testing the CIA mice serum TNF-a, IL-6, IL-17 expression level. Results C, D and B compared of TNF-a, IL-17 and IL-6 expression level decreased significantly; neutralizing anti-murine interleukin-17 antibody of the high concentration group were lower than low concentrations in serum inflammatory cytokine expression level, the difference was statistically significant (P <0.05). Conclusion Neutralizes interleukin-17 antibody can significantly reduce Inflammatory cytokine of the CIA mice model in serum TNF-a, IL-17, IL-6 expression level, And the highest concentration group improved joint function more significant than the low concentration group, The mechanism may be involved in regulating Th17 mediated by cytokines-osteoclasts immune system, Inhibit the inflammatory cytokine expression, Further affect osteoclast factor to achieve.
